Quench times are in seconds. B) Determination of Kdiss and kpol. CTP, ATP, and GTP were varied between 2 µM and 5 mM. Samples were quenched with HCl. Each data point indicates an apparent rate from exponential curve fitting (C12 + longer transcripts (product) versus time). Error bars indicate standard error from exponential fitting. C) Rpb2 R512C may have a defect in CTP-Mg2+ sequestration. At 5, 2.5, 0.5, and 0.1 mM CTP, reactions are quenched at 0.002 s with EDTA (E) or HCl (H).
