(A) Compensating mutations of the S. castellii Nvj1 ortholog. The N-terminal hydrophobic core sequence and flanking positive charges of the S. castellii ortholog can be altered to confer strict perinuclear localization in S. cerevisiae. EGFP reporters were localized were localized in nvj1Δvac8Δ and nvj1ΔVAC8+ cells. Nuclear chromatin (blue) and vacuole membranes (red) were co-stained with Hoechst and FM4-64, respectively. Interactions between Nvj1p-EGFP and Vac8p on FM4-64 stained vacuoles create NV junctions, which appear yellow in the overlay. Arrowheads in vac8-Δ cells point to cortical ER localization.
