Differentiated THP-1 cells were infected with M. tuberculosis H37Rv, the isogenic OppD-KO or OppD-KO complemented with oppDA at an MOI of 1:20 as described under “Materials and Methods” (A–C). After 24 h, cytokine release was measured in the supernatants using cytokine assay ELISA kits, or (D) cells were washed, lysed, and cell death was measured using the Cell Death ELISA kit (Roche) as described under “Materials and Methods”. Results of cell death assays are expressed as the fold increase in the release of histone compared to untreated THP-1 cells. Results represent the means ± S.D. of three determinations.
