Global telomeric G-overhangs undergo cell cycle-regulated dynamics regardless of telomerase expression. (AC) HeLa cells. (DF) IMR90 cells. (A, D) FACS analysis of DNA content in cells synchronized at the G1/S boundary and then released to S phase. Cells were collected at indicated time points for FACS analysis to determine the DNA content. A portion of IMR90 cells were permanently arrested at G0 and exited the cell cycle. (B, E) Measurement of G-overhang sizes using the overhang protection assay. The smears represent the heterogeneity of overhang sizes. ExoI digests 3′ overhangs and the ExoI plus (+) lanes show the background. The ∼54 nt band has been shown to be non-specific (Chai et al, 2005) and is not eliminated in the quantitations because its contribution to the overhang size is minimal.
