Effects of interleukin-1β (IL-1β) and interferon-γ (IFNγ) on cell viability in human CHME3 microglial cells treated with Aβ 142 (grey boxes) or vehicle (white boxes). The cells were preincubated with 50 ng/mL IL-1β and 50 ng/mL IFNγ for 24 hours prior to addition of Aβ 142 (1 μg/mL). At 24, 48, 72, and 96 hours after addition of Aβ 142 the viability of the cultures was assessed with the MTT assay. The data are expressed as % of control (vehicle) set at 100% and shown as median ± percentiles (25%–75% and 10%–90%), n = 4 (24, 48 h), n = 6 (72, 96 h). Statistical difference from vehicle is indicated by *(P < .05) and ***(P < .005), and statistical difference from Aβ 142 is indicated by #(P < .05) and ##(P < .01). Inline graphic(P < .01) indicates statistical difference between IFNγ and IL-1β + IFNγ, and (P < .05) indicate differences between IL-1β and IL-1β + IFNγ.
