Importance of the J domain HPD motif in Jac1 function. (A) 1:10 serial dilutions of each strain were spotted onto YP plates containing glucose and incubated at 30°C for 3 days, or at 23°C or 37°C for 4 days. (B) Mitochondria were prepared from strains expressing wild-type Jac1, Jac1 with a polyhistidine tag (Jac1His), or both the tagged Jac1 and the HPD mutant protein (Jac1A3 + Jac1His). Extracts were prepared and proteins were separated by SDS/PAGE. After transfer, the immobilized proteins were subjected to immunoblot analysis using either Jac1-specific antibodies (Left) or antibodies specific for the mitochondrial matrix protein Mge1.
