Reduced CO-difference spectrum of intact yeast cells harboring cDNA clone F14 (A). The smooth lower spectrum (B) illustrates the CO-difference spectrum of the corresponding isolated microsomes to demonstrate that the absorbance at 422 nm in (A) was due to an endogenous soluble yeast pigment. The microsomal preparation was catalytically functional in the NADPH and oxygen-dependent taxane 10β-hydroxylase reaction.
