Detection of hemifusion between viruses and target cells with a pH sensor. DiO-labeled (green) and CypHer5-labeled (red) viruses (mixing ratio 10:1) bearing SINmu (A) or HAmu (B) were prebound to 293T/CD20 cells for 30 min at room temperature. During live-cell imaging, virus-cell fusion was triggered by adding the appropriate volume of acidic buffer, pre-titrated to provide the desired pH (HAmu: pH 5.0, SINmu: pH 5.5), and monitored by pH drop (asterisk) and lipid transfer (arrow).
