Transfection efficacy (% of the cells transfected) and viability (those alive and adherent to the membrane) were measured following insonation with a 1 MHz pulse of 50 cycles at a PRF of 100 Hz while peak negative pressure was ramped from 0 to 950 kPa. Transfection efficacy was calculated as the percentage of fluorescent cells out of the total number of cells. Viability was calculated as the ratio of cell densities between cells treated with ultrasound and without ultrasound. Maximal transfection (0.87%) occurred at 300 kPa and corresponded to 24% viability (n=≥4, results expressed as mean +/− S.D., * indicates p < 0.05).
