Interaction of full-length htt with GST-CA150 fusion protein in vitro. Equivalent amounts of protein extracts from lymphoblastoid cells from three members of an HD-affected family were incubated with purified GST (lanes c) or GST-full length CA150 (lanes b). The input of protein extracts (lanes a) was also analyzed. Western blotting was performed with anti-htt mAb HU-4C8. Experiments were in triplicate. The intensity of HU-4C8 signal in lanes with GST-CA150 was similar in the three individuals tested, suggesting that CA150 binds equally to both normal and mutated full-length htt.
