By ELISA measurement, TNF-α (A) and IL-1β (B) levels were significantly increased in the spinal cord from SCI mice. On the contrary, olprinone treatment (0,2 mg/kg 1 h and 6 h after SCI induction) prevented the SCI-induced levels of these cytokines (A and B). In addition, spinal cord sections were processed at 24 h after SCI to determine the immunohistological staining for TNF-α and IL-1β expression. A substantial increase in TNF-α (D) and IL-1β (G) expression was found in inflammatory cells, in wm and gm of the spinal cord tissues from SCI mice. Tissue expression of TNF-α (E) and IL-1β (H) were significantly attenuated in olprinone -treated mice in comparison to SCI animals. The assay was carried out by using Optilab Graftek software on a Macintosh personal computer (CPU G3-266).
