RSCL-0520 inhibits TLR4-induced TNF-α secretion in THP-1 monocytes and PBMCs. THP-1 cells (A) and PBMC (B), 2 × 10⁵ cells per well, were pretreated with RSCL-0520 (50 µM) 1 h prior to TLR ligand treatment. The pretreated cells were stimulated with various TLR ligands at different concentrations (TLR1/2, 75 ng·mL⁻¹; TLR3, 75 µg·mL⁻¹; TLR4, 250 ng·mL⁻¹; TLR5, 75 ng·mL⁻¹; TLR6, 75 ng·mL⁻¹; TLR7/8, 7.5 µg·mL⁻¹; and TLR9, 7.5 µg·mL⁻¹) for 24 h according to the manufacturer's instructions. The culture supernatant was then assayed for TNF-α. Cells untreated with RSCL-0520 served as controls.
