Metformin suppression of the migration and invasion of human fibrosarcoma HT-1080 cells. (A) HT-1080 cells were incubated with varying concentrations of metformin in the presence of PMA (30 nM) or metformin only for 24 h in serum-free medium, and viability was determined by an MTT assay. Data are expressed as the means ± SD of triplicate experiments. *Significantly different from control (P < 0.01). (B) Cell monolayers were scratched with a pipette tip and then treated with metformin (1–5 mM) for 24 h. Migrating cells were photographed under phase contrast microscopy. (C) Cells were pretreated with metformin (5 mM) or vehicle (0.1% DMSO) followed by PMA (30 nM, 0.1% DMSO) treatment for 24 h. After 24 h, cells on the lower surface of the filter were counted.
