Synthesis and degradation rates of WT and nonmyristoylated c-Src. (A) COS-1 cells were transfected with the indicated c-Src constructs. At 22 to 24 h posttransfection, cells were pulse labeled for 15 to 20 min in DMEM containing Tran35S-labeled cysteine-methionine and chased in nonradioactive medium for the indicated times. At each time point, c-Src was immunoprecipitated from the cell lysates, and the amount of labeled c-Src was quantified and plotted relative to the amount of c-Src at time zero (100%). Average data for 2 independent experiments performed in duplicate are shown. (B) COS-1 cells transfected with the indicated c-Src constructs were pulse labeled with medium containing Tran35S-labeled cysteine-methionine for the indicated times, c-Src was immunoprecipitated, and the amount of labeled c-Src was quantified and plotted as a function of time. The inset shows autoradiographs for time points between 10 and 30 min. Representative data from experiments performed at least two times in duplicate are shown.
