(A) Transient expression of mRED-DN-myoVa and mRED control constructs in HeLa cells. HeLa cells were transfected with expression vectors for mRED-DN-myoVa-BRLT, mRED-DN-myoVa-MCLT, or mRED alone. Total cell lysates were collected at 0 and 20 hpi, separated on a polyacrylamide gel, and transferred to nitrocellulose. The blot was probed with a rabbit anti-RFP antibody. Signals from goat anti-lamin B antibody served as loading controls. (B) Myosin Va is required for efficient secretion of HSV-1 virions but not for intracellular production of infectivity. HeLa cells were transfected with expression vectors for mRED-DN-myoVa-BRLT, mRED-DN-myoVa-MCLT, or mRED alone. At 26 h posttransfection, cells were infected with HSV-1(F) at an MOI of 0.1. The resulting infectivity was determined by titration on Vero cells. The graph represents averages of data collected from three separate experiments.
