Protocol for preparation of sarcomeric subproteome using Triton X-100. This protocol was originally described by Solaro et al. (16). This method uses the non-ionic detergent Triton X-100 to solubilize the sarcolemmal, sarcoplasmic reticular (SR), and mitochondrial membranes, allowing the release of soluble proteins into solution and pelleting of the insoluble myofibrillar lattice in a rapid and efficient manner. All fractionation is performed in the presence of phosphatase and protease inhibitors to preserve the endogenous phosphostate and integrity of sarcomeric proteins. TnI, troponin I; TnT, troponin T.
