Comparison of reverse transcriptase product lengths. Copy number of one sequence near the polyA (1348–1412) and one near the 5′-UTR (22–80), respectively, were determined by qPCR. Full-length cDNAs act as templates for both qPCRs, while incomplete cDNAs only template replication from the PCR primer pair located near the polyA tail. The ratio of the two transcript represents the proportions of cDNA molecules that reached full-length (superscript III: 76%, superscript II: 70%, omniscript: 65%).
