Identification of actin as PX binding partner. Neutrophil cytosol was pre-absorbed with Glutathione-Sepharose^® 4B beads and then incubated with GST-p40PX-Glutathione-Sepharose^® 4B beads or Glutathione-Sepharose^® 4B beads as described in Section 2. Bound proteins were recovered and resolved by SDS-PAGE. The gel was stained with Coomassie blue R-250. Proteins marked by arrows were cut from the gel and digested in-gel by trypsin at 30 °C for 12 h. The first lane shows markers proteins with the molecular mass (kDa) indicated. The fourth lane shows the preparation of GST-p40PX used to immobilise.
