MiR-16 suppresses cell proliferation and sensitizes MCF-7 cells to doxorubicin. (A) Growth curves of Wip1+/+ and Wip1−/− MEFs infected with control or miR-16 expressing pseudoviruses. MCF-7 cells were transfected with control or miR-16 expression vector DNA. 36 h after transfection, cell lysates were harvested or total RNA was extracted for the analyses of miRNAs (by quantitative PCR, left panel) and proteins (by Western blotting, right panel). (C) MiR-16 sensitizes MCF-7 cells to doxorubicin. MFC-7 cells were transfected with control, Wip1 shRNA, miR-16 expression vector DNA, or miR-16 and Wip1 shRNA expression vector DNA. 48 h after transfection, cells were incubated with increasing doses of doxorubicin as indicated. Cell viability was determined 3 days after incubation.
