The synergistic effect of LCA on the cytotoxicity of TMZ depends on a functional pol β. Clonogenic survival of AAG-wild-type (A) and AAG-null (B) MEF cell lines modified to stably express exogenous wild-type or dominant negative (E295K-mutant) pol β, as indicated. Statistically significant differences at the p < 0.05 level (*) between treatment with 1 mM TMZ or 1 mM TMZ + 50 μM LCA were observed only in cells expressing wild-type pol β. The BER-deficient E295K mutants exhibited no significant difference in cytotoxicity between the treatments, indicating that LCA exerted no cytotoxic effects in the absence of wild-type pol β function.
