A model for the regulation of Myo5 recruitment and activation at the cortical endocytic patch. Cytosolic Myo5 interacts with Cmd1, which stabilizes a conformation where the Cext (C) folds back and binds to the TH1 (T) domain. Under this circumstance, the SH3 domain is not accessible for binding to Vrp1. An intracellular signal at the PM causes Cmd1 dissociation from the Myo5 neck and releases the inhibitory interaction. SH3-mediated binding of Myo5 to Vrp1 concentrates Myo5 at the cortical endocytic patches and triggers Myo5-induced actin polymerization. Cmd1 reassociation to the Myo5 neck contributes to dissociate it from the PM.
