WT skin has Smad2, -3, and -4 bound to the HGF promoter 466 bp upstream of the TSS, with the various Smads having recruited transcriptional activators (CBP/p300) and repressors (TGIF, CtBP1, and HDAC3). When Smad4 is lost, increased Smad2 is bound, recruiting transcriptional repressors and preventing HGF expression. In contrast, when Smad2 is lost, increased Smad3 and Smad4 bind to the HGF promoter, recruiting transcriptional activators and promoting HGF expression.
