Time course of dvrA mRNA levels in wild-type A. fumigatus (A) and the effect of dvrA disruption on germination (B). (A) Conidia of strain Af293 were incubated in Sabouraud broth at 37°C in a shaking incubator. At the indicated time points, an aliquot of the organisms was removed for RNA extraction. The dvrA transcript levels were determined by real-time PCR using tef1 as the endogenous control gene. Results are means ± standard deviations (SD) from two biological replicates, each measured in duplicate. (B) Conidia of the three strains of A. fumigatus were incubated in RPMI 1640 medium containing 50% fetal bovine serum at 37°C in 5% CO₂. At the indicated time points, the organisms were fixed and the percentage of cells that had germinated was determined. Results are means ± SD from three independent experiments.
