ICP0 expression restores the cytokine inhibitory activity of HSV mutant virus. HEK293 cells were transfected with TLR2, CD14, an NF-κB-driven firefly luciferase reporter gene, and a thymidine kinase (TK) promoter-driven Renilla luciferase reporter gene. Twenty-four hours later, the cells were infected with 7134 or n212 (two ICP0-deficent viral strains), the ICP0-rescued virus (7134R), or WT KOS. Luciferase activity was measured 6 h after viral infection and is expressed as firefly luciferase normalized to Renilla luciferase (or relative light units [RLU]).
