Effect of diguanylate cyclase protein expression on L. pneumophila biofilm formation. Growth of static cultures was monitored by measuring the optical density at 600 nm (OD 600). Biofilms were stained with crystal violet and resuspended in 95% ethanol before the absorbance at 600 nm (CV 600) was measured. A) The relative CV 600 and relative OD 600 of static cultures in 96 well polystyrene microtiter plates at 5 days post-inoculation for LP02 harboring pMMB206-GENT pvca0956 or plpg1057. B) 40x magnification images of post-exponential (OD 600 = 4.0) L. pneumophila grown in liquid cultures with shaking and 100 μM IPTG induction. LP02 harboring the pMMB206-GENT vector forms coccoid, motile cells in post-exponential phase, but Vca0956 and Lpg1057 induce filamentous morphology. **p-values<0.05 for CV 600 comparison of LP02 harboring the empty pMMB-206-GENT vector with LP02 harboring pvca0956 or plpg1057. *p-values<0.05 for OD 600 comparison of LP02 harboring the empty pMMB-206-GENT vector with LP02 harboring pvca0956 or plpg1057.
