Phosphorylation of PLD2 at Thr566 requires PLD2 localization to the lamellipodia of spreading cells. COS-7 cells were transfected with either a GFP control vector, GFP-PLD2WT, or the GFP-PLD2T566A or GFP-PLD2T566E mutant, as indicated. Cells were then serum starved for 24 h, detached from culture dishes, maintained in suspension for 3 h, and replated on FN. They were then fixed and stained with TRITC-labeled phalloidin.
