Effect of cortactin knockdown on p21WAF1/Cip1 and p27Kip1 mRNA levels. FaDu control and cortactin knockdown cells were synchronized into G1 phase by serum starvation and hydroxyurea (HU) treatment and then released into complete medium for the time periods indicated. Following this, mRNA was extracted from the cells, and qRT-PCR was performed to determine the relative amounts of p21WAF1/Cip1 (A) and p27Kip1 (B) mRNA in each sample, as described in Materials and Methods.
