A) representative FRET-images. CFP and YFP emission was detected by CLSM of transiently transfected mesophyll protoplasts. Emission was recorded in three channels comprising CFP-, FRET- and YFP-channel. CFP and YFP channels were essential for estimation of CFP-crosstalk and YFP-direct excitation. The average intensity was 2779 ± 953 in the YFP-channel, 1843 ± 982 in the CFP-channel and 3448 ± 650 in the FRET channel (mean ± SD, n=246).
