Depletion of CLOCK attenuates viral genes expression. HEp-2 cells were either mock transfected, transfected with the CLOCK siRNA, or the control siRNA for 80 h before infection with the HSV-1(F) (10 pfu/cell). The cells were harvested at 4 h, 9 h, or 24 h after infection. Lysates containing 60 μg of total proteins were separated on denaturing polyacrylamide gels, electrically transferred to nitrocellulose sheets, and immunoblotted for Clock, ICP4, ICP0, ICP8, U_S11, and β-actin, as described in Materials and Methods.
