ALP is inefficiently transported to the Golgi complex in erv26Δ cells. A) Wild-type (CBY2151) and erv26Δ (CBY2240) strains expressing full-length pro-ALP (pFL8) were monitored in pulse-chase experiments as described under Materials and Methods. Primary IPs were performed with anti-ALP or anti-CPY serums followed by secondary IPs to measure α1,6-mannose modified carbohydrate. Positions of pro-ALP (p), mature ALP (m) and a soluble breakdown product (*) are indicated as are the ER (p1), Golgi (p2) and mature (m) forms of CPY.
