The insertion of a spacer in the middle of the 116 nt alternative intron results in its accurate splicing out. (B) Analysis of pre-mRNA splicing of TPO cDNA+spacer constructs in Hep3B cells. Direct sequencing of the PCR products showed that the band corresponds to the TPO cDNA lacking 116 nt (993 bp).
