IL-2 overcomes iNKT cell anergy in vitro. Splenocytes from B6 and apoE−/− mice were labeled with CFSE and cultured for 24 hours with α-GalCer (100 ng/mL) with or without IL-2 (50 U/mL). The cells were then washed, and cultures were continued for an additional 72 hours without α-GalCer, in the presence or absence of IL-2. After culture, CFSE dilution was analyzed on iNKT cells.
