Specific knockdown of MK2 reduces stress-induced K20 phosphorylation. A, control shRNA or MK2 shRNA-transduced and selected HT29 cells were treated with 10 μg/ml anisomycin for 30 min. Lysates were analyzed by Western blotting using the indicated antibodies. Band intensities for pK20 and pHSPB1 were normalized to that of GAPDH to account for loading differences and plotted. B, control and MK2 shRNA-transduced HT29 cells were stimulated with anisomycin for 30 min or left untreated and then stained for K20-Ser(P)13.
