(A) Effect of ATP-depletion on 3H-3OC12-HSL levels associated with cells. A549 cells were ATP-depleted by incubation with 5 mM sodium azide and 80 mM 2-deoxy-D-glucose for 4 hours, while control cells received no treatment. Both groups were then exposed to 1.8nM 3H-3OC12-HSL and cells and cell associated radioactivity was assessed at the time points indicated. (B) 3H-3OC12-HSL continues to accumulate in cells treated with Glyburide. A549 cells were exposed to 250 μM Glyburide or the equivalent volume of DMSO as a vehicle control for one hour before being exposed to 1.8nM 3H-3OC12-HSL. Cell associated radioactivity was assessed at the time points indicated. The data are represented as the means±s.d.; n=3.
