Rate of reduction of bovine heart cytochrome c is generally correlated to the pKa of the amine in reaction with R5P. Lysine, however, shows an enhanced ability to generate products with R5P that reduce cytochrome c. A) Reduction of cytochrome c with amines of different pKa values. All reactions involved a 30-min pre-incubation of amine with R5P at 37 °C and pH 7.5 prior to addition to cytochrome c. Final reaction concentrations were: Amine (100 mM); R5P (10 mM); cytochrome c (1.0 mg/mL; 82 μM). Amines depicted on the graph were Lys (▲- - ▲); Gly-Gly (■ – ■); Arg (◆ – ◆); β-Ala (○ - - ○), and N-acetylLys (□ – □). The value for the lowest pKa contained by the molecule is listed on the figure.
