Treatment of abalone visceral extract enhances the cytolytic activity of CD8+ T cells. After 25 days of abalone visceral extract treatment, CD8+ T cells in tumor draining lymph nodes (axillary, superficial, brachial and linguinal lymph node) were isolated from each group. (A) Proliferative capacity of CD8+ T cells from each group was tested by [3H]-thymidine incorporation assay. (B and C) The expression levels of cytokines (TNF-α, IFN-γ) and cytolytic molecules such as granzyme B (GzmB), granzyme C (Gzmc) and FasL) were measured by quantitative real-time PCR. The mRNA expression levels in each group were normalized with HPRT (house-keeping gene) and then fold induction of each target gene was compared to PBS groups (expression level of control group: 1). (D) The cytotoxicity of CD8+ T cells was compared by JAM test. CD8+ T cells were co-cultured with [3H]-thymidine labeled EL4 T cells for 12 hours and the radioactivity in each sample was measured. Values are means with SE. One (*), two (**) or three asterisks (***) indicate p < 0.05, p < 0.01 or p < 0.001, respectively.
