(A) Immunocytochemical staining for sox2; (B) Western Blot analysis of sox2 expression in hAFS cells. Embryonic Carcinoma cell line NCCIT served as a positive control and human fibroblasts served as a negative control; (C) PCR analysis shows that undifferentiated hAFS cells express several cardiac markers including the transcription factors MEF2C and MEF2D, the gap junction protein connexin 43, and the surface proteins N- and H-cadherin. GAPDH served as an endogenous control. Human heart RNA served as a positive control while RNA without reverse transcription (RT) served as a negative control.
