The amidase-positive clone catalyzes the conversion of benzamide to benzoate (step 1). Benzoate activates the transcriptional regulator BenR, which in turn activates the benA promoter (PbenA) (step 2) and induces the expression of a reporter gene (gfp; step 3). Enzymatic benzoate production is measured as GFP fluorescence.
