Phosphomimetic mutation of Ser²⁶⁹ inhibits p53 growth-suppressive activity. H1299 cells were transfected with 1 μg of pcDNA plasmid containing wild type, S269A, or S269D. Twenty-four hours later, cells were trypsinized and diluted and then plated in medium containing 0.5 mg/ml Geneticin and grown for a further 2 weeks. Colonies were fixed and stained with Giemsa dye.
