Water (50 nl) containing 15 ng of cRNA of wild-type NKCC1 was injected on day 2. On day 3, 50 nl of water containing 10 ng of SPAK cRNA and 10 ng of WNK4 cRNA were injected. Bumetanide-sensitive K+ (filled bars) and Cl− (open bars) fluxes were measured on day 5 in hyperosmotic (265 mosmol l−1) conditions. Bars represent mean ±s.e.m. (n = 20–25 oocytes).
