HbpW1015A is accessible to protease digestion in the periplasm. Expression of Hbp and Hbp derivatives in MC1061degP::S210A cells was induced as described in the legend for Fig. 3. One hour after induction, cells were subjected to treatment with proteinase K. Proteins separated by SDS-PAGE were probed by immunoblotting with an antiserum against mature Hbp passenger (top panels) and with an antibody against the periplasmic chaperone SurA (bottom panels). The HbpW1015A mature form (m) and pro-form (p) are indicated at the right side of the panels; the digestion fragments of wild-type Hbp passenger are indicated with an asterisk.
