Model for the role and regulation of nucleotide synthesis during HR repair. Rad3 activation in response to a DSB leads to activation of the Ddb1–Cul4Cdt2 ubiquitin ligase complex. Complex activation results in Spd1 degradation, RNR activation, and subsequent increase in dNTP pools. This dNTP pool increase is required for postsynaptic gap filling during HR repair.
