(A) Sensitivity of ElpS-3Flag to proteinase K. Immunoblots with anti-Flag and anti-MreB antibodies were performed on total protein lysates harvested from cultures of CB15N elpS::elpS-3flag (WT elpS::elpS-3flag), CB15N Δ(gspC-gspN) elpS::elpS-3flag (Δ(C-N) elpS::elpS-3flag) grown in poor medium treated (+PK) or not (NT) with 2 µg/ml proteinase K. (B) Protease inhibitors sensitivity of ElpS-3Flag release in supernatant. Immunoblots with anti-Flag antibodies were performed on cellular extracts and precipitated supernatant from CB15N elpS::elpS-3flag grown in poor medium for 4.5 h then resuspended and incubated for 90 min in fresh medium containing (+ antiP) or not (-antiP) proteases inhibitors. Proteins contained in sample in (B) were detected by SDS-PAGE electrophoresis and silver nitrate staining as loading control for immunoblots.
