Human coronary artery smooth muscle cells (HCSMC) were transfected with an siRNA to decrease 11β-HSD1 expression, synchronized by serum starvation (0.2% serum) for 48 h, and stimulated with 5% fetal calf serum and platelet-derived growth factor-BB (PDGF) (10 ng/ml) in the presence or absence of dexamethasone (10−7 mol/L). Cell proliferation was determined by measuring BrdU incorporation (n=4).
