NF κB inhibitor suppressed H2O2-induced up-regulation of TLR10 expression. A representative RT-PCR data showing TLR10 expression in THP-1 cells were treated with TF inhibitors for 30 min or 1 h before treatment with 100 μM H2O2. AP-1, NF-AT and NF- κB were blocked with curcumin (Sigma, 10 μM, preincubation for 30 min), a cell-permeable NF-AT inhibitor peptide (Calbiochem, 4 μM, for 1 h) and quinazoline (Enzo Life Sciences Inc., 10 μM, for 30 min), respectively. The cells that had been maintained at 37 °C in 20% O2 in the presence or absence of the inhibitors were treated with 100 μM H2O2 for 1 h, harvested and then examined by RT-PCR analysis. The relative expression of TLR10 was normalized by GAPDH using densitometric analysis. *P < 0.05.
