EcoRII translocation process analyzed by HS-AFM. Four frames out of 40 frames total taken over a time period of 20 s measured in 0.5 s intervals are shown. In the first frame (5s) looped structure stabilized by the protein (indicated with an arrow) bound to two specific sites on DNA is formed. The following frames illustrate sliding of the protein along the DNA strand leading to an increase in the loop size. The final image (20 s) in which a large loop is formed corresponds to binding of the protein to two distant binding sites on the DNA substrate.
