FIGURE 1.

Antiretroviral activity of C-A1. A, HIV-1 vector and HIV-1 LAI Δenv (VSV-G pseudotyped) virus were tested in SupT1 cells in single cycle assays; HIV-1 NL4.3 was tested in C8166 cells. Infection efficiency was measured by flow cytometry to detect GFP+ cells (HIV-1 vector and HIV-1 LAI Δenv) 24–48 h post-infection or by p24 immunostaining (HIV-1 NL4.3) 72 h post-infection. IC50, dose required to inhibit infection by 50%. CC50, dose inducing death of 50% of the cells. Mean values (n = 2) or mean values ± S.D. (n = 3) are shown. B, chemical structure of novobiocin and C-A1. C, PBMCs infected with HIV-1 SF-162 in the presence of C-A1 and analyzed 72 h post-infection by p24 capsid immunostaining. D, PBMCs infected with three HIV-1 primary isolates in the presence of C-A1 and analyzed 72 h post-infection by p24 CA immunostaining. NI, could not be infected; ND, not done.
