Effect of C2-ceramide on SK1a and SK1b expression. Western blot showing the effect of (a) C2 ceramide (50 μm, 24 h) on SK1a and SK1b levels in LNCaP-AI cells; (b) reversal by MG132 (10 μm, 30 min pretreatment) of the C2-ceramide (50 μm, 24 h)-induced reduction in SK1b levels in LNCaP cells and SK1a in hPASMC. The Western blots were reprobed with anti-ERK-2 antibody to ensure comparable protein loading. Results are representative of three separate experiments.
