Gal-1-9-1 and gal-1L9 form higher order multimers. (A) BS³ crosslinking of constructs reveals that gal-1L9 forms multimers up to pentamers, whereas gal-1-9-1 forms dimers and trimers. Proteins were incubated with crosslink reagent or buffer control and analyzed by SDS–PAGE. (B) Multimerization of galectins occurs under native conditions and does not occur through the CRD-active site. Proteins in 50 mM lactose or buffer control were run on a 4–12% Bis–Tris native gel.
