The kinetic isotope effect on flavin reduction by [4S-2H]-NADPH was determined in the reductive half-reaction using the labelled substrate in stopped-flow experiments. Anaerobic Lot6p (final concentration of 15 µM) was mixed with 200 µM anaerobic NADPH (dashed curve) or 200 µM [4S-2H]-NADPH (solid curve) in 100 mM CHES, pH 9.7 at 4 °C. Reaction traces (456 nm) are shown on a logarithmic time scale.
